Custom Antibodies - Monoclonal Antibodies Production

The Monoclonal Antibody Facility is pleased to offer:

• Monoclonal antibody production in mice and rats
• Conjugation to carrier proteins
• No limit on the number of clones provided
• Antibody quantification and sub-class determination
• New culture room and equipment

The Facility integrates extensive knowledge in the production of monoclonal antibodies against a variety of antigens, such as peptides, purified proteins, whole cells, membranes, carbohydrates, small molecules, etc. The Facility staff will invite the clients to discuss their needs and the applications for which they wish to use the antibodies in order to establish strategic immunization and screening protocols to select the hybridomas of choice.

Standard hybridoma service
The standard monoclonal antibody service includes:

• Antigen emulsion in Titermax adjuvant
• Injection of at least 3 animals
• Sera testing
• Choice of screening method (ELISA, Western blot, flow cytometry, others)
• Multiple (up to 3) antigen screening (ELISA)
• Cloning, sub-cloning and cryopreservation
• 10 vials of frozen cells for each positive clone obtained. For convenience, 5 vials are sent to the client and 5 vials are kept at the BRI Facility
• 100 ml of culture supernatant of each positive clone for further characterization

Options

Consultation
Clients can benefit from consultation and support services regarding:

• Troubleshooting in all aspects of hybridoma establishment
• Assistance in antibody characterization
• Development of hybridoma screening methods

Immunoglobulins characterization
The Custom Antibody Facility can offer additional characterization services such as:

• Antibody sub-class determination
• Quantification

Frozen hybridoma cell bank hosting
Cryopreservation and cryostorage of hybridoma cell lines can be provided by the BRI Custom Antibody Facility on an individual charge basis.

Training
The BRI Custom Antibody Facility can provide training of laboratory personnel for hybridomas production on an individual charge basis.

Complementary work
The Custom Peptide Facility can also assist in the design, synthesis and conjugation of peptide antigens. As well, the HTS Facility could be used for high throughput screening of hybridoma supernatant.

Animal Care Standard Procedures
All protocols used by the BRI Custom Antibody Facility are in accordance with the Canadian Council on Animal Care guidelines.
